| Medium for isolation of pure cultures
To isolate pure cultures, light-emitting pinpoints are transferred to agarmedium by means of sterile toothpicks. The medium has the following composition:
Tryptone (10 g), yeast extract (5 g), glycerol (3 ml), salt (20 g), magnesium sulfate (1 g), Tris-buffer (6 g), agar (20 g) dissolved in 1 liter of water. The pH is adjusted to 7.5, and the medium autoclaved at 121 °C for 30 minutes.
